Document Description
The California Department of Fish and Wildlife is issuing a permit to Aspen Workman (Permit No. 2081(a)-23-001-RP) pursuant to Fish and Game Code section 2081(a) for a project to improve the population genetic understanding of Tidestrom’s lupine. Results are expected to inform future habitat management decisions to conserve the genetic integrity of Tidestrom’s lupine. Plant tissue collection activities will take place between March and April of 2023. Three to five leaves will be collected from fifteen to twenty individuals at each location. At five sites where Tidestrom’s lupine co-occurs with Chamisso bush lupine (Lupinus chamissonis), putative hybrids will also be sampled totaling up to twenty leaf tissue collections per site. Leaves will be pinched off the stem at the base of the petiole, placed in envelopes and stored on silica gel until DNA extraction activities can be performed at Helmoltz Center for Environmental Research GmbH- UFZ in Germany. Each individual plant will be photographed, and GPS coordinates will be recorded. Data on reproductive status and morphology of the plant may be recorded to inform hybridization analyses. Plant tissue DNA will be extracted using a Qiagen DNEasy plant kit and samples will be processed using sequence-capture methodology. Following analysis, sequence data will be uploaded to public repositories.

Collections will also take place at Point Reyes National Seashore (38.0785, -122.9730) in Marin County, CA and Sonoma Coast State Park (38.4473, -123.1260) in Sonoma County, CA.
