Document Description
The California Department of Fish and Wildlife is issuing a permit to Benjamin Carter [Permit No. 2081(a)-21-013-RP] pursuant to Fish and Game Code section 2081(a) for a project to quantify Erysimum genetic diversity within and among geographically disjunct populations of E. menziesii and E. capitatum var. angustatum to reconstruct phylogenetic understanding of the genus. Primary research goals are to: (1) determine if disjunct populations of E. menziesii and E. capitatum var. angustatum are genetically distinct; and (2) determine evolutionary relationships within Erysimum. A small amount of leaf tissue (approximately 2 cm2) will be collected from 15 individual plants at each of the five sampling locations providing for a total of 75 samples for population genetic analysis material. One voucher specimen will be collected from each of the five populations, totaling five voucher specimens. Project activities include DNA extraction of collected plant tissue, polymerase chain reaction (PCR) amplification and genome sequencing to provide population statistical data for phylogenetic research purposes.

Plant material collection for the Project will occur in MacKerricher State Park (39.4932N, 123.7936W) in Mendocino County, Asilomar State Beach (36.6180N, 121.9419W) and Marina State Beach (36.6963N, 121.8091W) in Monterey County, Samoa Dunes (40.7627N, 124.2216W) in Humboldt County and Antioch Dunes in Contra Costa County (38.0160N, 121.7960W). DNA sequencing and molecular lab work will be conducted at the QB3 Vincent J. Coates Genomics Sequencing Laboratory, University of California Berkeley in Alameda County. Voucher specimens will be housed in the Carl W. Sharsmith Herbarium, San Jose State University, Santa Clara County.

Reasons for Exemption
The project consists of basic data collection and research that will facilitate the genetic understanding of two California endangered species, E. menziesii and E. capitatum var. angustatum. The permit issued by the California Department of Fish and Wildlife for the Project includes measures to ensure that existing populations of E. menziesii and E. capitatum var. angustatum are not detrimentally impacted by research activities.
